No. Toughening changes the stress in the glass, not its density, so a toughened pane weighs the same as an ordinary one of the same size.
Calculate each pane separately and add them; the gas in the cavity is negligible against the glass. The frame and spacer are not included either.
The arithmetic is exact — volume times density. The density is where the uncertainty sits, and the source behind this calculator carries acrylic exactly while glass is quoted as a range.
